    How OPDIVO Works

    1

OPDIVO works by blocking PD-1 (Programmed Death-1), a checkpoint protein on T-cells that normally acts as a "brake" on the immune system. Cancer cells exploit this pathway by expressing PD-L1 and PD-L2, which bind to PD-1 and suppress T-cell activity, allowing tumors to evade immune destruction.

    2

By blocking PD-1, OPDIVO releases this brake and restores the T-cell's ability to recognize and kill cancer cells. This approach harnesses the body's natural immune defenses rather than using cytotoxic agents that damage both cancer and healthy cells.

    3

When combined with YERVOY (ipilimumab), which blocks a different checkpoint called CTLA-4, the dual checkpoint blockade provides complementary mechanisms of immune activation, often resulting in higher response rates than either agent alone.
